I'd recommend entering via Chokoloskee if you want good fishing.

There is the big Park map - pdf that shows chickee and ground campsites.
If time and weather allow you might do a loop of both the outside islands (Pavillion Key/Rabbit Key) and the interior chickees (Sunday Bay/Sweetwater)
Chokoloskee Charters (dot com)can shuttle you deep via boat to a chickee and you can work your way back at a more relaxed pace that allows fishing.
Google Earth is a super fun way to devise a route.
Briefly, bring a spinning rod and some soft plastic lures/jigs/ spoons. If the weather is cold fish in the deeper water of the rivers or perhaps the very shallow bays as the sun warms them up. Casting the shorelines is always fun and often productive. Really cold weather can slow things down. Look for moving water and clear water. It's all so dang fishy looking it will drive you nuts.
